Increased Antimutagenic and in Vitro Anticancer Effects of Black Soybean (Seoritae) Chungkukjang Fermented by Bacillus subtilis

|
Black soybean (Seoritae) chungkukjang was fermented with different kind of starters (Bacillus subtilis KCCM 11315, Bacillus licheniformis CN-115 and Bacillus megaterium SMY-212). Antimutagenic and in vitro anticancer effects of the black soybean chungkukjangs were studied by Ames test using Salmonella enterica serovar Typhimurium TA100 and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay on AGS human gastric adenocarcinoma cells and HT-29 human colon adenocarcinoma cells, respectively. The NH2-N and NH3-N contents in the black soybean chungkukjang fermented with Bacillus subtilis (BS-C) were similar levels in the soybean chungkukjang fermented with Bacillus licheniformis (BL-C). Methanol extracts from the black soybean chungkukjangs showed higher antimutagenicities
against aflatoxin B1 (AFB1) and N-methyl-N¡¯-nitro-N-nitrosoguanidine (MNNG). At the 2.5 mg/plate concentration, the BS-C exhibited 79% antimutagenicity against AFB1, while the black soybean chungkukjang fermented with Bacillus megaterium (BM-C) revealed 33% antimutagenicity. The BS-C and BL-C also increased antimutagenic effect against MNNG than BM-C. The BS-C and BL-C showed higher inhibitory effects on the growth of AGS human gastric adenocarcinoma cells than the BM-C at the same concentration. And the BS-C exhibited the highest inhibitory effects (76%) on the growth of HT-29 human colon adenocarcinoma cells among the fermented chungkukjangs (p£¼0.05). In this study, black soybean chungkukjang fermented with different kind of starters exerted differently on the antimutagenic and
anticancer activities of the chungkukjangs. B. subtilis-fermented chungkukjang showed the highest chemopreventive
